Metaxalone is a high-dose oral solid, and that changes the questions worth asking a supplier. When a single tablet carries several hundred milligrams of API, the powder’s physical behaviour — bulk density, flow, particle size — stops being a footnote and starts driving whether your tablet press runs at all.

What Metaxalone is used for

Metaxalone is a centrally acting skeletal muscle relaxant, formulated for the relief of discomfort associated with acute, painful musculoskeletal conditions, typically alongside rest and physical therapy. It is an oral solid, most commonly in 400 mg and 800 mg tablets.

For a formulator, the high unit dose is the defining constraint. There is little room for bulky excipients, so the API’s own compressibility and flow have to carry more of the formulation than they would for a low-dose molecule.

Bulk density and flow are part of the specification

At 400 or 800 mg per tablet, the powder characteristics of the incoming API determine tablet weight, hardness and press throughput. Ask the supplier for bulk and tapped density and, where relevant, particle-size distribution, and treat these as release-relevant attributes rather than nice-to-have data.

A lot that passes identity and assay but flows poorly or packs to a different density can still disrupt a validated compression process. For a high-dose tablet, the physical specification is the part most likely to explain a batch that runs on the press one week and fights it the next.

Grade selection that matches your filing

Confirm which monograph or in-house specification the quoted material is controlled to, because that determines the analytical evidence you release against. A price quoted without a stated grade is not comparable to one that is, and this is the most common reason two quotations look far apart.

Ask, too, whether the physical attributes you rely on are held as controlled limits or merely reported as typical values. Only a committed limit protects your compression process from lot-to-lot variation, so it is worth pinning down before you validate around a single sample.

Packaging, storage and documentation before the first PO

Confirm the pack sizes, liner configuration and storage conditions in writing, and specify the pack size at enquiry stage if your warehouse decants — repacking after despatch adds a handling step your QA team has to document. Agree storage conditions that protect the material against light and moisture as the specification requires.

A batch Certificate of Analysis, the method of analysis, an MSDS and the product specification should be available before you order, with DMF or open-part documentation on request where your filing needs it. How quickly a supplier sends this set at enquiry stage tends to predict how they will respond when you have a shipment held at a port.

A short checklist before you commit

  • Ask for bulk and tapped density, and particle-size data where relevant.
  • Confirm whether physical attributes are controlled limits or typical values.
  • Confirm the pharmacopoeial grade the price is quoted against.
  • Agree pack size and liner configuration in writing.
  • Request a batch CoA and method of analysis before raising the PO.
