Pregabalin API: uses, applications and what to check on the specification sheet

A plain-language guide for formulators and procurement teams evaluating Pregabalin suppliers — indications, grade differences, impurity considerations and the documentation to ask for.

Pregabalin is one of the most widely formulated oral solid APIs in the Indian market. That popularity is exactly why supplier selection is harder than it looks: the material is available from many sources, and the differences that matter are not visible on a price list.

What Pregabalin is used for

Pregabalin is a gamma-aminobutyric acid analogue formulated principally for neuropathic pain — diabetic peripheral neuropathy and post-herpetic neuralgia — as an adjunct therapy in partial onset seizures, and in some markets for generalised anxiety disorder.

For a formulator, the practical consequence is that the same API feeds several dossiers with different regional monograph references. A supplier who can cover IP, BP, USP and EP from one route saves you qualifying two sources.

Grade differences that actually change your filing

IP, BP, USP and EP monographs differ in limits and in the analytical methods used to demonstrate them. The material is not different by grade so much as the evidence required to release it is.

Ask which monograph a quoted price is against. A price quoted without a stated grade is not comparable to one that is, and this is the single most common reason two quotations look far apart.

Impurity profile and route consistency

Because impurities arise from the synthetic route, a supplier who changes route between campaigns hands you a new impurity profile to justify. That is a regulatory problem long before it is a quality problem.

The question to ask is not “what is your impurity profile” but “has your route changed in the last three years, and will it change during our contract”.

Packaging and storage in practice

Standard packing is HDPE drums with double LDPE liners in 1 kg, 5 kg and 25 kg configurations. Pregabalin should be stored in tightly closed containers, protected from light and moisture at controlled room temperature.

If your warehouse handles smaller decants, specify the pack size at enquiry stage — repacking after despatch introduces a handling step your QA team will have to document.

Documentation to request before the first PO

A batch Certificate of Analysis, MSDS, product specification and method of analysis should be available before you order, not after. DMF and open-part documentation should be available on request where your filing needs it.

How quickly a supplier sends this set is a reliable early signal. Documentation turnaround at enquiry stage tends to predict documentation turnaround when you have a shipment held at a port.

If a supplier cannot send you a batch CoA and a method of analysis before you raise a purchase order, you are not evaluating a manufacturer.

• Confirm the pharmacopoeial grade the price is quoted against.
• Ask for a batch CoA and method of analysis before raising the PO.
• Confirm whether the synthetic route has changed recently.
• Agree the pack size and liner configuration in writing.
• Establish who prepares export documentation, and how long it takes.
